In most cases, specific media formulations are required for different species and different media are sometimes used within a species. However, the majority of culture media contain: water, macronutrients, micronutrients, vitamins, sugars, organic supplements, support media, osmoticum, and hormones.
Culture temperatures of 25-28 degrees Celsius are commonly used. Usually the cultures are grown in the dark. In addition, there are optimal plating densities that vary among species.
In anther culture, both solid and liquid media can be used. The results obtained are slightly different for each of these types of media; more calli/embryoids are produced when using liquid cultures but solid media often produces better quality embryoids.
